Target Information

Glutamate is the main excitatory neurotransmitter in the brain. Traditionally, it was known to act on ligand-gated receptor channels such as NMDA, AMPA, and kainate receptors, which mediate fast excitatory synaptic transmission. More recently, glutamate has also been found to regulate enzymes producing second messengers via G-protein–coupled receptors known as metabotropic glutamate receptors (mGluRs).
Group-I receptors (including mGluR1a and mGluR5) stimulate phospholipase C activity, leading to increased phosphoinositide turnover and calcium release from internal stores.
Group-II and Group-III receptors (e.g., mGluR2, mGluR3) are coupled to inhibition of adenylyl cyclase.
