Target Information
Glutamate is the main excitatory neurotransmitter in the brain. It was traditionally thought to act only on ligand-gated receptor channels (NMDA, AMPA, and kainate receptors) involved in fast excitatory synaptic transmission. Recent studies have shown that glutamate also regulates enzymes producing second messengers via specific G-protein-coupled receptors, known as metabotropic glutamate receptors.
Group-I receptors (mGluR1a and mGluR5) stimulate phospholipase C, leading to increased phosphoinositide turnover and calcium release from internal stores. Group-II and Group-III receptors (including mGluR2 and mGluR3) are coupled to inhibition of adenylyl cyclase.
