Product Specific Information

The LN3 monoclonal antibody reacts with human MHC class II molecule HLA-DR, expressed on antigen presenting cells (B cells, monocytes, macrophages, dendritic cells, and activated T cells).
HLA-DR plays a key role in peptide presentation to CD4+ T lymphocytes.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Pre-diluted and tested on normal human peripheral blood cells. Recommended usage is 5 µL (0.03 µg) per test (100 µL final volume).
• Excitation / Emission: 405 nm / 702 nm (Violet laser)
• Recommended Filter: 710/50 bandpass filter
• Light Sensitivity: Protect from light (photo-induced oxidation sensitive)
• Fixation Compatibility: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer or 1-step Fix/Lyse Solution with minimal brightness loss.
• Buffer Recommendation: Use Super Bright Complete Staining Buffer (Product # SB-4401) when combining multiple Super Bright dyes to minimize non-specific polymer interactions.

Target Information

HLA-DR is a transmembrane glycoprotein composed of a 36 kDa alpha chain (DRA) and a 27 kDa beta chain (DRB).

• The alpha chain gene (DRA) has 5 exons encoding the leader peptide, extracellular domains, transmembrane, and cytoplasmic regions.
• DRA acts as the sole alpha chain for DRB1, DRB3, DRB4, and DRB5.
• The beta chain (DRB) contains polymorphisms determining peptide binding specificity.
• Hundreds of DRB1 alleles are known and used in transplantation typing.
• Expressed primarily on antigen presenting cells such as B cells, monocytes, macrophages, thymic epithelial cells, and activated T lymphocytes.
• HLA and MHC antibodies are critical in Immunopeptidomics for neoantigen identification via LC-MS/MS.
