Target Information

Survivin (IAP4) is the smallest member of the Inhibitors of Apoptosis Protein (IAP) family. IAPs are involved in multiple cellular functions such as signaling, division, and metabolism, and show differential expression in nearly all human cancers but not in most normal tissues.

Survivin contains a single baculovirus IAP repeat (BIR) domain and lacks a C-terminal RING finger domain. It is highly expressed during fetal development and in most tumors, but low in adult tissues. Survivin expression is cell-cycle regulated, peaking in the G2/M phase, where it associates with mitotic spindle microtubules.

Disruption of survivin–microtubule interactions leads to loss of anti-apoptotic function and increased caspase-3 activity during mitosis. Survivin is also expressed in brain tissues (astrocytes and some neurons) of adult rats following traumatic brain injury, co-expressed with NeuN and PCNA, and may influence neural cell proliferation after injury.

At least four transcript variants exist, with three full-length isoforms identified. Overexpression of survivin in cancer may override apoptotic checkpoints and promote abnormal mitotic progression.
