Target Information
The 6x His tag is a synthetic oligopeptide consisting of six consecutive histidine residues (HHHHHH). It is commonly expressed at either the N- or C-terminal regions of recombinant proteins to enable isolation or purification via immobilized metal affinity chromatography (IMAC).
Epitope tagging allows detection of proteins for which no specific antibody exists by fusing a known epitope to the recombinant protein. His-tagged proteins are purified based on the strong and selective affinity of histidine clusters to metal ions such as Ni²⁺, enabling purification to homogeneity using Ni²⁺-NTA adsorbents.
His tag-specific antibodies, such as HIS.H8, facilitate detection and co-immunoprecipitation of tagged proteins.
