Caspase 3 is a cysteine-aspartic acid protease that interacts with Survivin, XIAP, CFLAR, Caspase 8, HCLS1, Deleted in Colorectal Cancer, TRAF3, and GroEL.
Located at chromosome 4q35, this gene encodes a key enzyme in the execution phase of apoptosis. Caspases are synthesized as inactive proenzymes and activated by proteolytic cleavage. Caspase 3 cleaves and activates caspases 6, 7, and 9, and is itself processed by caspases 8, 9, and 10.
It is the predominant caspase involved in cleavage of amyloid-beta precursor protein, linked to neuronal death in Alzheimer’s disease. Caspase-3 activation in heart failure generates truncated SRF, functioning as a dominant-negative transcription factor.
Additionally, caspase-3 influences bone mineral density and plays a role in erythroid precursor differentiation, where Hsp70 prevents CASP3-mediated cleavage of GATA1 and apoptosis.
