Target Information
The TEK receptor tyrosine kinase is expressed almost exclusively in endothelial cells. TEK signaling plays a critical role in endothelial cell–smooth muscle cell communication during venous morphogenesis. Mutations in TEK are associated with inherited venous malformations. The ligand angiopoietin-1 binds to TEK and mediates signaling essential for embryonic vascular development. TIE2 expression is upregulated by thyroid-stimulating hormone and agents that increase intracellular cAMP. Hematopoietic stem cells (HSCs) expressing TIE2 are quiescent, antiapoptotic, and adhere to osteoblasts in the bone marrow niche.
