The monoclonal antibody 4-29-5-10-21 reacts with human and non-human primate CD43 (leukosialin, galactoglycoprotein, sialophorin). CD43 is a sialomucin with both adhesive and anti-adhesive functions, expressed on most leukocytes and a small subset of B lymphocytes. Proteolytic processing upon activation decreases surface expression.

CD43 is involved in activation of T cells, B cells, NK cells, and monocytes. The counter-receptor for CD43 is CD169/SIGLEC-1, expressed on macrophages.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal rhesus peripheral blood cells. Use at 5 µL (0.06 µg) per test (100 µL final volume). Recommended cell number: 10⁵–10⁸ cells/test.

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm). Use 710/50 bandpass filter (alternative: 695/40). Ensure instrument compatibility.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal brightness impact.

Target Information

CD43 (leukosialin, sialophorin) is a transmembrane mucin-like protein with high negative charge, expressed on most hematopoietic cells. It contributes to a repulsive barrier interfering with adhesion, but can promote leukocyte aggregation. By interacting with ezrin and moesin, CD43 regulates T-cell morphology and lymphocyte traffic.

CD43 signaling enhances LFA-1 adhesiveness and modulates other receptor pathways. Expression induces tumor suppressor p53, but in p53/ARF deficiency, promotes tumor proliferation. CD43 dysfunction is associated with Wiscott-Aldrich Syndrome and Adenoid Basal Cell Carcinoma.
