Product Specific Information

Antibody 32-5800 reacts specifically with human PTEN protein. Reactivity was confirmed with cell lines containing endogenous PTEN (NIH3T3, K652, DU145) and transfected cell lines (Rat1, 293T). Detects ~55 kDa PTEN protein and is effective in western blot analysis of breast cancer cell lines. No cross-reactivity to other proteins observed.

Target Information

PTEN is a tumor suppressor gene frequently mutated in various cancers. It encodes a phosphatidylinositol-3,4,5-trisphosphate 3-phosphatase with both tensin-like and catalytic domains similar to dual specificity tyrosine phosphatases. PTEN dephosphorylates phosphoinositide substrates, negatively regulating intracellular PI(3,4,5)P3 levels and the AKT/PKB signaling pathway. It plays a key role in AKT-mTOR signaling, influencing neuron integration, dendritic development, and synapse formation. PTEN also negatively regulates insulin signaling and glucose metabolism in adipose tissue. Nuclear monoubiquitinated PTEN exhibits higher apoptotic potential, while cytoplasmic nonubiquitinated PTEN shows reduced tumor suppressive activity.
