Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. p53 upregulation triggers the transcription of genes such as p21waf1, leading to G1 or G2/M arrest.

The structure of p53 includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerization domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is critical for p53 activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 plays a key role in preventing proliferation of damaged cells, contributing to its anticancer function. It is also implicated in aging, oxidative stress, and cognitive processes such as long-term potentiation (LTP).

Mutations in TP53 gene are common in human cancers, leading to loss of tumor suppressor activity. Both somatic and germline mutations (e.g., in Li-Fraumeni syndrome) are known to disrupt normal p53 function.
