Product Specific Information

The HI30 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells excluding circulating erythrocytes and platelets. The cytoplasmic portion of CD45 has tyrosine phosphatase activity and plays an important role in lymphocyte activation.

• Applications Tested: Pre-diluted and tested on normal human peripheral blood cells.
  Recommended use: 5 µL (0.25 µg) per test in a final volume of 100 µL.
  Cell number range: 10⁵–10⁸ cells/test.

Alexa Fluor 561 emits at 575 nm and is ideal for use on spectral cytometers, allowing multiplexing with PE and PE-eFluor 610.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase (PTP) expressed on all nucleated hematopoietic cells, comprising ~10% of lymphocyte surface proteins. It is absent on non-hematopoietic cells and tissues. CD45 plays a key role in lymphocyte development and antigen signaling by regulating Src-family kinases. Multiple isoforms exist due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions. CD45 regulates T- and B-cell receptor signaling, suppresses JAK kinases, and modulates cytokine receptor signaling, integrin-mediated pathways, and apoptosis-related functions.
