Product Specific Information
The MEL-14 monoclonal antibody reacts with mouse CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells, binding glycosylated, fucosylated, and sulfated sialylated glycoproteins such as CD34, GlyCAM-1, and MAdCAM-1. These interactions mediate lymphocyte rolling on activated endothelium and homing to high endothelial venules (HEV) of peripheral lymphoid tissues.
Applications Reported:
This MEL-14 antibody is validated for use in flow cytometric analysis.
Performance Notes:
Internal testing indicates Alexa Fluor 561 may non-specifically stain B cells in Swiss Webster and SJL mice, but not in BALB/c or C57BL/6 mice. Other strains have not been tested.
Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended usage ≤ 0.25 µg per test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best performance.
Fluorophore Characteristics:
Alexa Fluor 561 emits at 575 nm and is suitable for spectral cytometers, multiplexed with PE and PE-eFluor 610.

Target Information
Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor with a C-type lectin domain, EGF-like domain, two SCR sequences, a spacer region, transmembrane region, and short cytoplasmic tail. It is constitutively expressed on most leukocytes including lymphocytes, monocytes, and polymorphonuclear cells, except for a subset of memory T cells.
