Product Specific Information

Description:
The RPA-T4 monoclonal antibody reacts with human CD4, a 59 kDa cell surface receptor expressed by most thymocytes, a subpopulation of mature T-helper cells, and at low levels on monocytes. CD4 is a receptor for HIV. RPA-T4 blocks HIV binding and mixed lymphocyte reaction. The RPA-T4 antibody recognizes a different epitope than the OKT4 monoclonal antibody and does not cross-block binding.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.25 µg) per test (final volume 100 µL; 10⁵–10⁸ cells/test).

Light Sensitivity:
PE-Cyanine5 is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples may be stored in IC Fixation Buffer (Product #00-8222-49) or 1-Step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal effect on brightness.

Excitation/Emission:
488–561 nm / 667 nm
Laser: Blue, Green, Yellow-Green
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD4 antigen is involved in MHC class II recognition and serves as a co-receptor for HIV. It is expressed mainly on T-helper lymphocytes and at lower levels on monocytes, macrophages, and dendritic cells. CD4 expression is also found in thymus, lymph nodes, tonsils, spleen, and some non-lymphoid tissues. It plays a key role in T-cell activation and may mediate neuronal damage in immune-related diseases.
