Product Specific Information

  • Recommended positive controls: HCT116 cells, or HCT116 treated with 30 µM cisplatin for 24 hours.
  • Predicted reactivity: Mouse (100%), Rat (100%), Bovine (100%).
  • IHC notes: Requires antigen retrieval using heat-mediated 10 mM Citrate buffer (pH 6.0) or Tris-EDTA buffer (pH 8.0).
  • Store as a concentrated solution. Centrifuge briefly before opening the vial.

Target Information

CHK1 (CHEK1) is a serine/threonine protein kinase that prevents DNA replication when genomic integrity is compromised. It plays a central role in the DNA damage-induced checkpoint network and is evolutionarily conserved.
When CHK1 function is inhibited, p53-deficient cancer cells undergo mitotic catastrophe and apoptosis, while normal cells arrest in G1 phase. Therefore, CHK1 inhibition can enhance the efficacy of DNA-damaging agents in cancer therapy, making CHK1 an attractive target for anti-cancer research.
