Target Information

The insulin-like growth factor-binding proteins (IGFBPs) are a family of homologous proteins that have co-evolved with the IGFs. They act as shuttle molecules for soluble IGFs and regulate IGF signaling. IGFBPs influence IGF bioavailability, concentration, and extracellular distribution. Additionally, they have biological activities independent of IGFs.
Seven IGFBPs have been identified, each differing in tissue distribution, half-lives, and modulation of IGF receptor interactions.

  • IGFBP1: Negatively regulated by insulin; highly expressed during fetal liver development and under nutritional or diabetic conditions.
  • IGFBP2: Functions as a chaperone escorting IGFs to target tissues; expressed in fetal eye and brain.
  • IGFBP3: Most abundant; complexes with ~80% of serum IGFs; released by dermal fibroblasts upon incision injury.
  • IGFBP4: Also released by fibroblasts after injury.
  • IGFBP5: Secreted by myoblasts; may play a key role in muscle differentiation.
