Target Information
Phosphoenolpyruvate Carboxykinase 1 (PCK1)는 포도당신생합성(gluconeogenesis)의 주요 조절 효소로, 이 과정의 속도 제한 단계(rate-limiting step)를 담당합니다.
PCK1은 gluconeogenesis를 통해 CH8(+) T 세포의 기억 형성과 유지에 관여하며, 시트르산 회로 내 대사 중간체의 수준을 cataplerosis와 anaplerosis 조절을 통해 제어합니다.
- 고혈당 상태: PCK1은 옥살로아세트산(oxaloacetate)을 포스포엔올피루브산(PEP)으로 전환합니다.
- 저혈당 상태: PCK1은 반대로 PEP을 옥살로아세트산으로 전환합니다.
- Ser-90 인산화 시: AKT1에 의해 인산화되면, PCK1은 단백질 키나아제 활성을 가지며, INSIG-SCAP 상호작용을 방해하고 SREBP 단백질의 핵 내 이동을 촉진하여 지방생성 관련 유전자 발현을 조절합니다.
PCK1의 주요 상동 유전자는 PCK2입니다.
