Product Specific Information

The monoclonal antibody A1exF5 recognizes both human and mouse Arginase 1 (Arg1), a cytosolic enzyme.
It is compatible with standard intracellular staining protocols and the Foxp3/Transcription Factor Staining Buffer Set.

• Applications Reported: Intracellular staining followed by flow cytometric analysis.
• Applications Tested: Tested on normal human lysed whole blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00).
• Recommended ≤ 1.0 µg per test (100 µL final volume).
• Cell number range: 10⁵–10⁸ cells/test.
• Antibody titration recommended for optimal performance.

Brilliant Violet™ 711 (BV711) emits at 713 nm and is suitable for cytometers equipped with a violet (405 nm) laser.
Use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific polymer interactions when using multiple polymer dye-conjugated antibodies.

Light Sensitivity: Protect from light to avoid photo-induced oxidation.
Fixation: Compatible with IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark.
Not compatible with methanol-based fixation.

Target Information

Arginase-1 (Arg1) is a 35 kDa enzyme that converts L-arginine to urea and L-ornithine, the final step in the urea cycle.
It regulates nitric oxide production by depleting arginine, the substrate for NO synthase.
Arginase 1 is expressed in liver, neutrophils, myeloid-derived suppressor cells (MDSC), and neural stem cells.
In mice, it is a hallmark of alternatively activated macrophages (M2a).
Defects in Arginase 1 cause argininemia, an autosomal recessive disorder characterized by hyperammonemia.
