Product Specific Information

The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by a majority of thymocytes, T helper cells, and low levels on monocytes. CD4 acts as a receptor for HIV, and SK3 blocks HIV binding and mixed lymphocyte reaction. SK3 and RPA-T4 antibodies recognize distinct epitopes.

Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for human peripheral blood cells at 5 µL (0.06 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which provides improved formulation over previous versions. Add 5 µL per sample for optimal staining in antibody cocktails or single-color controls.

NovaFluor conjugates are based on Phiton™ technology, utilizing novel nucleic acid dye structures engineered for minimal spillover and spread effects.

Target Information

CD4 antigen is involved in MHC class II molecule recognition and serves as a co-receptor for HIV. It is primarily expressed in T helper cells, as well as monocytes, macrophages, and dendritic cells. CD4 is found in thymus, lymph nodes, tonsils, spleen, and certain regions of brain and gut. It plays a role in early T-cell activation and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
