Description:
The M18/2 monoclonal antibody reacts with mouse CD18, a 90–95 kDa integrin beta 2 glycoprotein. CD18 is the common beta subunit that associates with CD11a (alpha L), CD11b (alpha M), and CD11c (alpha X) to form LFA-1, Mac-1, and gp150/95. These heterodimers are expressed broadly by leukocytes, with NK and T cells showing higher expression. CD18 is involved in cellular adhesion and immune response through interactions with counter-receptors.

Applications Reported:
Suitable for flow cytometry, immunoprecipitation, immunoblotting (WB), and immunohistochemistry. Also used in blocking CD18 in functional assays. (For functional assays, use Functional Grade purified M18/2, Product #16-0181).

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes and bone marrow cells. Recommended ≤0.5 µg per test. A test is defined as the amount of antibody that stains a cell sample in 100 µL volume. Cell number may range from 10⁵ to 10⁸ cells/test. Antibody titration is recommended for optimal performance.

Target Information

CD18 (integrin beta 2) is a 90 kDa type I transmembrane protein expressed on all leukocytes. It forms heterodimers with CD11a–d to constitute leukocyte (β2) integrins such as LFA-1 (CD11a/CD18), Mac-1 (CD11b/CD18), and others. These integrins are essential for leukocyte migration and intercellular adhesion. Deficiency or mutation in CD18 leads to leukocyte adhesion deficiency type I (LAD1) and related immune dysfunctions. CD18 also serves as a receptor for Mannheimia haemolytica leukotoxin.
