CDK4/cyclin D3 serine/threonine kinase는 세포 주기의 G1 단계에서 세포 증식 조절에 관여하는 이합체 단백질 복합체입니다.
CDK4는 cyclin D형 단백질과 복합체를 형성하며, p16(cyclin-dependent kinase inhibitor-2)에 의해 억제됩니다.
CDK4는 Rb 단백질의 C-말단 영역을 인산화하여 E2F 복합체의 전사 억제 활성화에 기여할 수 있습니다.
CDC37과 HSP90은 D형 cyclin에 결합하지 않은 CDK4 분획과 선택적으로 결합할 수 있습니다.
SMAD3는 CDK4 및 CDK2의 주요 생리적 기질입니다.
CDK6과 CDK4 모두 Rb 단백질을 인산화할 수 있으나, 서로 다른 잔기 선택성을 보입니다. (CDK6: Thr821, CDK4: Thr826)
