Target Information
Tau는 신경세포 축삭에서 주로 발견되는 미세소관 결합 단백질로, tubulin 중합을 촉진하고 미세소관을 안정화하는 기능을 가집니다.
C-말단은 축삭 미세소관에 결합하고 N-말단은 신경 세포막 구성 요소에 결합하여 Tau가 두 구조 사이의 연결 역할을 수행합니다.
Tau의 과인산화 형태는 알츠하이머병(AD) 뇌에서 신경섬유 엉킴의 구성 요소인 paired helical filaments(PHF)의 주요 구성 요소입니다.
과인산화는 Tau의 미세소관 결합 기능을 저해하여 미세소관 불안정을 초래하고, 결국 신경세포의 퇴화를 유발합니다.
Tau는 GSK-3β, PKA, cdk5, casein kinase II 등 다양한 세린/트레오닌 키나아제에 의해 인산화됩니다.
Hyper-phosphorylated Tau는 Pick병, 전두측두엽 치매, corticobasal degeneration, progressive supranuclear palsy 등 다양한 중추신경계 질환에서도 발견됩니다.
