Product Specific Information
The MAb11 antibody reacts with human tumor necrosis factor-alpha (TNF alpha), a 17 kDa cytokine produced by monocytes, macrophages, neutrophils, NK cells, and CD4+ T cells. TNF alpha exhibits cytolytic activity against various tumor cells and plays an important role in immune regulation. It exists as soluble dimers and trimers or as a 26 kDa membrane-bound form.
MAb11 is a neutralizing antibody suitable for intracellular staining followed by flow cytometric analysis. Tested using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00) at 5 µL (0.125 µg) per test. A test is defined as staining a sample in 100 µL final volume (typically 10^5–10^8 cells/test).
Alexa Fluor 660 emits at 690 nm and is ideal for spectral cytometry multiplexing with APC, Alexa Fluor 647, and Alexa Fluor 700.

Target Information
TNF alpha is a multifunctional proinflammatory cytokine belonging to the TNF superfamily. Secreted mainly by macrophages, it binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR.
It regulates immune responses, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. TNF alpha forms multimers (dimers, trimers, pentamers) and shows a single 17 kDa band under non-reducing SDS-PAGE.
Mouse knockout studies suggest a neuroprotective role. TNF alpha induces tumor necrosis and participates in immune responses against bacterial, viral, parasitic, and fungal infections. It acts synergistically with interferon-gamma in cytotoxicity and is implicated in autoimmune diseases, insulin resistance, and cancer.
