Product Specific Information
Description:
The 4S.B3 monoclonal antibody reacts with interferon-gamma (IFNγ). Human IFNγ is a 17 kDa cytokine produced by activated T and NK cells with antiviral and antiparasitic activity. It acts synergistically with TNFα to inhibit proliferation of normal and transformed cells and modulates immune responses on various cell types expressing IFNγ receptors. Glycosylation does not affect its biological activity.
Applications Reported:
Suitable for intracellular staining followed by flow cytometric analysis.
Applications Tested:
Pre-diluted and tested for intracellular staining of stimulated human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).
Recommended usage: 5 µL (0.015 µg) per test in 100 µL final volume. Cell number may vary from 10⁵ to 10⁸ cells/test.
Fluorophore Characteristics:
Alexa Fluor 660 emits at 690 nm and is suitable for use on spectral cytometers.

Target Information
IFN gamma (Interferon gamma, Type II interferon):
A macrophage activation factor produced mainly by T-lymphocytes and NK cells in response to antigens and cytokines. It is a dimeric glycoprotein (~45 kDa) composed of two 146 amino acid subunits. Functions include antiviral and antiproliferative activity, induction of MHC I/II, macrophage activation, and enhanced immunoglobulin secretion.
Human IFNγ does not cross-react with mouse IFNγ. Activation occurs via IFNγ receptor I/II and the JAK-STAT pathway. Expression is upregulated by IL-2, FGF basic, EGF and downregulated by vitamin D3 or DMN. Mutations are associated with aplastic anemia.
