Product Specific Information

The MP6-XT22 antibody reacts with mouse tumor necrosis factor-alpha (TNF alpha), a 17 kDa cytokine produced by monocytes, macrophages, neutrophils, NK cells, and CD4(+) T cells. TNF alpha has cytolytic activity against various tumor cells and plays a critical role in immune regulation. It exists as soluble dimers/trimers and a 26 kDa membrane-bound form.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested on stimulated mouse splenocytes using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and protocol. Recommended usage ≤ 0.25 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Titrate for optimal performance.

Alexa Fluor 660 emits at 690 nm and is suitable for spectral cytometers.

Target Information

TNF alpha is a multifunctional proinflammatory cytokine of the TNF superfamily, secreted mainly by macrophages. It binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR receptors and regulates immune cell activity, apoptosis, lipid metabolism, and coagulation. It appears as a 17 kDa band under non-reducing SDS-PAGE and forms multimers in solution. TNF alpha plays roles in immune defense, tumor necrosis, and multiple diseases including autoimmune disorders, insulin resistance, and cancer.
