Product Specific Information

The M1/69 monoclonal antibody reacts with the mouse CD24 molecule, also known as Heat Stable Antigen (HSA).
This 35–50 kDa GPI-anchored membrane protein is expressed by erythrocytes, thymocytes, peripheral lymphocytes, and myeloid lineage cells.
CD24 expression varies by lineage due to glycosylation heterogeneity and is useful for identifying B-cell development stages in mouse bone marrow. It is also known to interact with P-selectin (CD62P).

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes; recommended ≤0.25 µg/test (100 µL final volume). Optimize per assay.

Fluorochrome Information:
Super Bright 600 is a tandem dye excited with a 405 nm violet laser and emits at 600 nm. Recommended detection using a 610/20 bandpass filter.

Staining Recommendations:
When using multiple Super Bright conjugates, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Handling Notes:

• Light sensitivity: Protect from light; dye is photo-sensitive.
• Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333).
• Laser: Violet (405 nm); Emission: 600 nm.

Target Information

CD24 is a GPI-anchored glycoprotein expressed at multiple stages of B-cell development, from pro-B cells to mature surface Ig-positive B cells.
Expression is low or absent in plasma cells. CD24 is also found on most B-lineage acute lymphoblastic leukemias, B-cell CCLs, and B-cell non-Hodgkin’s lymphomas.
It plays a role in B-cell proliferation, maturation, and autoimmunity regulation.
