PEP-043 is a 15 amino acid synthetic peptide whose sequence corresponds to residues 2016-2027 of human SMRT and an N-terminal addition of C G G as a linker for peptide conjugation to a carrier protein. The sequence of this peptide is (amino to carboxy terminus): C-G-G-A2016-S-A-S-D-P-H-R-E-K-T-Q2027

This peptide may be used for neutralization and control experiments with the monoclonal antibody that reacts with this product and SMRT, catalog # MA1-842 & MA1-843. Using a solution of peptide of equal volume and concentration to the corresponding antibody will yield a large molar excess of peptide (~70-fold) for competitive inhibition of antibody-protein binding reactions.

Steroid and thyroid hormones and retinoic acid regulate a complex array of gene expression activity via intracellular receptor transcription factors belonging to the ligand-dependent nuclear receptor superfamily. Adding to the complexity of function of these transcription factors are associated proteins known as coactivators and corepressors which, as their names suggest, enhance or depress transcription activity of the nuclear receptor with which they associate. Silencing mediator of retinoic acid and thyroid hormone receptor (SMRT) and nuclear receptor corepressor (N-CoR) are related transcriptional corepressors which contain two distinct domains capable of interacting with unliganded nuclear receptors to repress their basal transcriptional activity.
