The 6B11 monoclonal antibody recognizes an epitope of the CDR3 formed by the germ-line configuration of the V alpha 24 and J alpha 18 of the TCR alpha locus, resulting in the expression of an invariant T-cell receptor (TCR) expressed by invariant NK T cells (iNKT). This human TCR is homologous to the murine V alpha 14 J alpha 18 and pairs predominantly with V beta 11 chains.

NK T cells are restricted by the CD1d molecule and activated by CD1d-presented glycolipid ligands, leading to rapid production of cytokines such as IL-4, IL-13, and IFN-γ. Although iNKT cells have NK-like cytolytic activity, they act as regulators of immune responses by producing both Th1 and Th2 cytokines in autoimmune, infectious, and cancer contexts. The proportion of iNKT cells in normal human peripheral blood varies between 0.01–1% of total T cells.

Target Information

T cell receptors (TCRs) enable discrimination between self and foreign peptides presented by MHC class II molecules, essential for adaptive immunity. Recognition of self-peptides is linked to autoimmune diseases, while mutant self-peptides are associated with tumors. Superantigen engagement of TCRs can cause toxic shock syndrome. Autoantibodies to TCR V beta segments have been found in rheumatoid arthritis and SLE, potentially modulating inflammatory responses. Most human T cells express TCR alpha-beta with CD4 or CD8, while a small double-negative subset is found in certain autoimmune and immunodeficiency disorders. Gamma-delta T cells, mainly in epithelia, exhibit distinct antigen recognition and immunoregulatory roles.
