• For immunohistochemistry (PFA fixed), incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD81 (TAPA-1, target of anti-proliferative antibody-1) is a member of the tetraspanin family, expressed on most nucleated cells, particularly germinal center B cells.
It forms complexes with other tetraspanins, integrins, coreceptors, and MHC molecules, influencing cell adhesion, morphology, activation, proliferation, and differentiation.
In muscle, CD81 promotes cell fusion and myotube maintenance, and it also serves as a receptor for the hepatitis C virus.
Like other tetraspanins (CD9, CD37, CD53, CD63, CD82), CD81 has four hydrophobic domains and mediates signal transduction involved in cell development and motility.
CD81 complexes with CD19, CD21, Leu 13, and integrins, and antibodies against CD81 show anti-proliferative effects on lymphoid cell lines, particularly large cell lymphomas.
It is localized in a tumor-suppressor gene region and is considered a candidate gene for malignancies.
