Target Information

T-cell protein tyrosine phosphatase (TC-PTP), also known as PTPT and PTPN2, is an enzyme that removes phosphate groups covalently attached to tyrosine residues in proteins. This enzyme exists in two C-terminal splice variants with distinct subcellular localizations:

• 45 kDa isoform: Exclusively nuclear in resting cells, redistributes to the cytosol upon stimulation with growth factors and cellular stress.
• 48 kDa isoform: Found in the endoplasmic reticulum and has different physiological substrates.

TC-PTP is prominent in hemopoietic cell types. Knockout mice lacking TC-PTP die 3–5 weeks after birth due to erythropoietic and lymphopoietic deficits, indicating its critical role in bone marrow maturation. It dephosphorylates various phosphoproteins, including p52 Shc and receptors for EGF, Insulin, and growth hormone.
