Target Information
The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, crucial for reproductive processes such as pregnancy establishment and maintenance.
PR exists as two forms: 94 kDa (Form A) and 120 kDa (Form B), generated by alternative translation initiation. PR-B is transcriptionally active, regulating genes for endometrial maintenance, pregnancy, and ovulation inhibition.
PR-A lacks 165 amino acids at the N-terminus, exposing an inhibitory domain that represses transcriptional activity.
In its inactive state, PgR forms a multiprotein complex with heat shock proteins and immunophilins. Progesterone binding induces conformational change, receptor dimerization, and DNA binding to progesterone response elements (PRE), activating transcription.
A null mutation in the PGR gene results in reproductive abnormalities.
