Target Information
Adhesion to the extracellular matrix regulates cell survival through integrin engagement and cell spreading. Anoikis is apoptosis induced by integrin detachment. Amino-terminal enhancer of split (AES) is part of the Groucho/transducin-like enhancer of split (TLE) family of transcriptional co-repressors, involved in neurogenesis, segmentation, and sex determination. AES forms a complex with Bit1 (Bcl-2 inhibitor of transcription 1), a mitochondrial protein released during apoptosis. This complex may suppress survival-promoting gene transcription regulated by TLE proteins. Apoptosis of AES and Bit1-transfected cells can be inhibited via attachment to fibronectin through α5β1 integrin, indicating that the Bit1-AES pathway contributing to anoikis is integrin-regulated, particularly by α5β1 integrin.
