Product Specific Information

The 4H11 monoclonal antibody reacts with human CD34, also known as mucosialin. CD34 is a single-pass transmembrane protein with a heavily glycosylated extracellular and N-terminal mucin domain. It is expressed on hematopoietic progenitors, mesenchymal stem cells, tumor cell lines, and vascular endothelia in adults.

Epitopes of CD34 have been classified into three classes (I, II, III) based on enzymatic cleavage sensitivity. The 4H11 antibody belongs to class III, indicating reactivity with a protein epitope.

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometry of normal human peripheral blood cells (≤0.5 µg/test, in 100 µL final volume). Cell count should be empirically determined (10⁵–10⁸ cells/test). Titrate for optimal performance.

Target Information

CD34 is a highly glycosylated surface protein (111–115 kDa) expressed on stem cell populations. It serves as a stem cell marker and may function in receptor-mediated endocytosis, adhesion, differentiation, and proliferation of hematopoietic stem cells. CD34 expression reflects specific stages of hematopoietic development and may mediate cell attachment to the bone marrow matrix or stromal cells.

CD34 possibly acts as a scaffold for lineage-specific glycans, facilitating interactions with lectins. The intracellular domain is phosphorylated by protein kinase C, suggesting a role in signal transduction.
Diseases associated with CD34 include dermatofibrosarcoma and neurofibroma.
