Target Information

RNA polymerase III synthesizes tRNA, 5S rRNA, 7SL RNA, and U6 snRNA. It is overexpressed in many transformed cell lines and tumors, as cells must duplicate its protein components before division. High levels of Pol III transcribed RNA are required for rapid growth, involving TFIIIB and TFIIIC2 transcription factor complexes.
TFIIIC2 consists of TFIIIC220, TFIIIC110, TFIIIC102, TFIIIC90, and TFIIIC63 subunits, which are overexpressed in adenovirus-transformed and malignant cells such as ovarian carcinomas. TFIIIC2 recruits RNA pol III and TFIIIB to promoter elements and may play a key role in deregulation of malignant cells.
The TFIIIB complex includes TBP (TATA-binding protein), BRF1 (TFIIIB90), and TFIIIB. Expression of these factors is upregulated in transformed cells. In many carcinomas, tumor suppressors RB and p53 are inactivated, affecting their ability to bind and inactivate TFIIIB function.
