Target Information
Oxygen radicals damage chromosomal DNA, causing cell death and inducing mutations. Although much focus has been on the repair of 8-oxo-G, many other DNA lesions arise from free-radical attacks. The human NTH1 repair protein acts on a wide range of oxidatively damaged DNA sites. As a homologue of E. coli Endonuclease III, NTH is a DNA glycosylase with apurinic/apyrimidinic lyase activity. It shows broad substrate specificity, including ring-saturated and fragmented pyrimidine products. Research suggests NTH plays a crucial role in removing oxidative base lesions in mitochondrial DNA.
