Target Information
Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, upon binding to estrogen, undergoes conformational change enabling dimerization and DNA binding to estrogen response elements (ERE). ERs regulate gene transcription via coactivator or corepressor recruitment.
There are two forms of the estrogen receptor: alpha (ESR1) and beta (ESR2), encoded by separate genes. Alternative RNA splicing produces multiple alpha isoforms (66, 53, 47, and 35 kDa). ERs are expressed in various tissues and are essential for sexual development, reproductive function, and bone health. They are implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
