Target Information
Ubiquitin exists either covalently attached to another protein or free (unanchored). When covalently bound, it is conjugated to target proteins via an isopeptide bond as monoubiquitin or polyubiquitin chains linked through different lysine residues or via the initiator methionine.
Each linkage type has distinct biological roles:
- Lys-6-linked: DNA repair
- Lys-11-linked: ERAD and cell-cycle regulation
- Lys-29-linked: Lysosomal degradation
- Lys-33-linked: Kinase modification
- Lys-48-linked: Proteasomal degradation
- Lys-63-linked: Endocytosis, DNA-damage response, NF-κB signaling
- Linear (Met-linked): Cell signaling
