Product Specific Information

The B3B4 monoclonal antibody reacts with mouse CD23, a 45 kDa type II transmembrane glycoprotein expressed on resting conventional B cells. Expression is modulated upon B-cell activation, while B-1 lineage (CD5+ B cells) does not express CD23. Soluble CD23 forms are biologically active and function as low affinity IgE receptors involved in IgE regulation and B-cell activation. CD21 interacts with CD23.

• Applications Tested: Tested on mouse splenocytes at ≤ 0.5 µg/test (100 µL volume, 10⁵–10⁸ cells/test). Optimize titration for best performance.
• Fluorochrome: Super Bright 600 (excitation: 405 nm, emission: 600 nm). Use 610/20 bandpass filter.
• Staining Buffer: When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.
• Light Sensitivity: Protect from light to prevent photo-oxidation.
• Fixation: Samples may be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal signal loss.

Target Information

CD23 is a 45 kDa glycoprotein expressed on subsets of peripheral blood and tonsil B cells, and highly expressed on EBV-transformed B lymphoblasts. It functions as a low-affinity IgE receptor and plays roles in B cell growth, differentiation, and IgE production. Soluble CD23 acts as a potent mitogenic factor. CD23 expression is associated with certain B cell malignancies including chronic lymphocytic leukemia and centroblastic/centrocytic lymphoma.
