Product Specific Information

The HB5 monoclonal antibody reacts with human CD21, an approximately 145 kDa type I transmembrane protein expressed by mature B cells, follicular dendritic cells, and a subset of epithelial cells. CD21 serves as a receptor for complement component C3d and Epstein-Barr virus (EBV).
CD21, in association with CD19 and CD81, forms a multimolecular complex with BCR and plays a role in B cell activation.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated for flow cytometry using normal human peripheral blood cells. Recommended usage: 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Recommended detection filter: 610/20 bandpass.

Usage Notes:

• When using multiple Super Bright-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.
• Protect from light to avoid photo-induced oxidation.
• Fixed samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C with minimal impact on brightness and FRET efficiency.

Target Information

CD21 (complement receptor 2, CR2, C3D receptor, EBV receptor) binds complement fragments such as C3d and plays crucial roles in:

• Uptake and retention of immune complexes
• Survival of memory B cells
• Development and maintenance of humoral immune responses
• EBV binding and prion targeting to follicular dendritic cells

Soluble CD21 (sCD21) retains ligand-binding capability. CD21 interacts with CD23 and associates with CD19, CD81, and Leu13 to form a signal-transduction complex for B cell activation.
Genetic variations in CD21 are associated with susceptibility to systemic lupus erythematosus type 9 (SLEB9).
Alternative splicing generates different isoforms.
