Product Specific Information
The 61D3 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, interfollicular macrophages, and some dendritic cells. CD14 binds LPS-LBP complexes with high affinity on monocyte surfaces.
Each product includes:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated on normal human peripheral blood cells at 5 µL (0.6 µg)/test.
A test is defined as staining a cell sample (10⁵–10⁸ cells) in 100 µL final volume.
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
- Add 5 µL per sample before combining with cells.
- For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).
Technology:
NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures engineered for minimal spectral spillover and spread impact.
Excitation: 494 nm; Emission: 585 nm; Laser: 488 nm (Blue).
Target Information
CD14 is a 55 kDa GPI-anchored glycoprotein expressed on monocytes, macrophages, and neutrophils. It functions as a lipopolysaccharide receptor and exists in both membrane-bound and soluble forms. CD14 interacts with LPS-binding protein (LBP) and plays roles in immune recognition, inflammation, and apoptosis.
Associated diseases include Mycobacterium chelonae infection and Croup.
Alternative splicing produces multiple transcript variants of the same isoform.
