Target Information
SMAD2 (MADH2, MAD2)는 세포 증식, 세포사멸, 분화 등 다양한 세포 과정을 조절합니다.
SMAD2는 TGF-beta 수용체와 상호작용하여 신호 전달을 매개하며, 핵 내로의 이동은 TGF-beta 신호 전달의 핵심 단계입니다.
ERK1에 의한 MH1 도메인의 칼모듈린 결합 영역 내 Thr8 인산화는 SMAD2의 전사 활성화를 증가시키며, 이는 칼모듈린에 의해 음성 조절됩니다.
TGF-beta 신호에 반응하여 SMAD2는 TGF-beta 수용체에 의해 인산화되고, SARA로부터 분리되어 SMAD4와 결합합니다.
SMAD4와의 결합은 핵 내 이동 및 전사 억제 복합체 형성에 중요합니다.
또한 SMAD2는 activin type 1 receptor kinase에 의해 인산화되어 activin 신호 전달에도 관여합니다.
대체 스플라이싱된 SMAD2 단백질 변이체도 보고되었습니다.
