The Ras superfamily of small GTP-binding proteins are key mediators in cell signaling pathways regulating proliferation, cytoskeletal organization, and secretion. Their activity is modulated by GTPase regulators: guanine nucleotide-releasing factors (GRFs) that increase GDP dissociation, and GDP-dissociation inhibitors (GDIs) that decrease it.
The Rho GDI subfamily includes Rho GDIα, Ly-GDI (Rho GDIβ), and Rho GDIγ, which interact with Ras-like GTP-binding proteins such as Rho A, Rho B, Rac, and Cdc42. Ly-GDI is expressed mainly in hematopoietic cells, particularly B and T lymphocytes.
