Target Information
DNA-directed RNA polymerase II subunit RPB1 (POLR1A) is a DNA-dependent RNA polymerase that catalyzes the transcription of DNA into RNA using the four ribonucleoside triphosphates as substrates.
POLR1A is the largest subunit and a catalytic component of RNA polymerase II, which synthesizes mRNA precursors and various functional non-coding RNAs. It forms the polymerase active center with the second largest subunit.
Pol II is the central component of the basal RNA polymerase II transcription machinery, composed of mobile elements that move relative to each other. RPB1 is part of the core element with a central large cleft, a clamp element that opens and closes the cleft, and jaws that grab the incoming DNA template.
At transcription initiation, a single-stranded DNA template strand of the promoter is positioned within the central active site cleft of Pol II. A bridging helix emanates from RPB1 and crosses the cleft near the catalytic site, promoting translocation of Pol II by acting as a ratchet mechanism.
During elongation, Pol II moves along the template as the transcript elongates. This process is regulated by phosphorylation of the C-terminal domain (CTD) of RPB1, which serves as a scaffold for factors controlling transcription initiation, elongation, termination, and mRNA processing. Gene expression levels are influenced by the balance of methylation and acetylation of CTD lysine residues.
