Target Information
Immunophilins are a family of soluble cytosolic receptors that bind to immunosuppressant agents cyclosporin A (CsA) or FK506. Proteins binding FK506 are termed FK506 Binding Proteins (FKBPs), while those binding cyclosporin A are called cyclophilins (CyP). Both CyP:CsA and FKBP:FK506 complexes inhibit calcineurin, a calcium- and calmodulin-dependent phosphatase involved in T-cell activation, providing a mechanism for immunosuppression.
Immunophilins act as peptidyl prolyl cis-trans-isomerases (PPIase), accelerating protein folding in vivo and in vitro. Within the cyclophilin family, CyPA, CyPB, and CyPC share high homology. CyPA (also known as CyP-18) is the most abundant and ubiquitous cyclophilin found in all vertebrate tissues and is present in T-cells.
