Target Information
The expression of DUSP1 gene is induced in human skin fibroblasts by oxidative or heat stress and growth factors. It encodes a protein similar in structure to non-receptor-type protein-tyrosine phosphatases and shares significant sequence similarity with a Tyr/Ser-protein phosphatase encoded by the vaccinia virus H1 gene. The bacterially expressed and purified DUSP1 protein exhibits intrinsic phosphatase activity, specifically inactivating MAP kinase in vitro by dephosphorylating both phosphothreonine and phosphotyrosine residues. It also suppresses MAP kinase activation by oncogenic ras in Xenopus oocyte extracts. Thus, DUSP1 may play an important role in the cellular response to environmental stress and in the negative regulation of cell proliferation.
