Target Information

CUL4B is a core component of multiple cullin-RING-based E3 ubiquitin-protein ligase complexes, mediating ubiquitination and proteasomal degradation of target proteins. It acts as a scaffold protein, positioning substrates and ubiquitin-conjugating enzymes for catalysis.
CUL4B participates in polyubiquitination of CDT1, histones H2A, H3, and H4 in response to DNA damage, and is targeted to UV-damaged chromatin by DDB2, playing a role in DNA repair and replication. It is required for cyclin E ubiquitination and normal G1 cell cycle progression, and regulates the mTOR pathway controlling cell growth and metabolism. Specific regulation of the mTORC1 pathway by CUL4B depends on 26S proteasome function and interaction with MLST8.
