Target Information

CUL4B is a core component of multiple cullin-RING-based E3 ubiquitin-protein ligase complexes that mediate ubiquitination and subsequent proteasomal degradation of target proteins.
The functional specificity of the E3 ubiquitin-protein ligase complex depends on the variable substrate recognition subunit.
CUL4B may act as a scaffold protein, positioning the substrate and ubiquitin-conjugating enzyme for catalysis.
It plays a role in polyubiquitination of CDT1, histone H2A, H3, and H4 in response to radiation-induced DNA damage.
CUL4B is targeted to UV-damaged chromatin by DDB2 and is important for DNA repair and replication.
It is required for ubiquitination of cyclin E and normal G1 cell cycle progression.
Additionally, CUL4B regulates the mTOR pathway involved in cell growth and metabolism, dependent on 26S proteasome function and interaction with MLST8.
