Target Information

CUL4B is a core component of multiple cullin-RING-based E3 ubiquitin-protein ligase complexes that mediate ubiquitination and proteasomal degradation of target proteins. It acts as a scaffold protein, positioning substrates and ubiquitin-conjugating enzymes for catalysis.
CUL4B participates in polyubiquitination of CDT1 and histones H2A, H3, and H4 during radiation-induced DNA damage response. It is targeted to UV-damaged chromatin by DDB2 and plays a role in DNA repair and replication.
Additionally, CUL4B is required for cyclin E ubiquitination and normal G1 cell cycle progression, and it regulates the mTOR pathway controlling cell growth and metabolism. This regulation depends on 26S proteasome function and interaction between CUL4B and MLST8.
