Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 up-regulates stress response genes such as p21waf1, leading to G1 or G2/M arrest.

Structurally, p53 includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerization domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is crucial for activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 plays key roles in tumor suppression, aging, oxidative stress, and memory-related processes. Mutations in TP53 are linked to various cancers and hereditary syndromes such as Li-Fraumeni syndrome.
