Product Specific Information
- Tested in direct ELISA (Use antibody at 2–4 µg/mL for coating).
- For Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C followed by cooling at room temperature for 20 min.
Target Information
CD22 (BL-CAM) is a type 1 integral membrane glycoprotein (130–140 kDa) expressed in both cytoplasm and membrane of B-lymphocytes.
It appears early in B-cell differentiation and is useful for phenotyping mature leukemias. CD22 is strongly expressed in hairy cell leukemia and is positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.
CD22 preferentially binds to α2,6-linked sialic acid and plays a regulatory role in B-cell receptor signaling through interactions with Src family tyrosine kinases and cytoplasmic phosphatases.
