Product Specific Information
- Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
CD22 (BL-CAM) is a type I integral membrane glycoprotein (130–140 kDa) expressed in the cytoplasm and membrane of B-lymphocytes.
It appears early in B-cell differentiation, similar to CD19, and is expressed in mature B cells but absent in plasma cells.
CD22 expression is useful for phenotyping mature leukemias and is strongly expressed in hairy cell leukemia.
It binds preferentially to α2,6-linked sialic acid, regulating B-cell receptor signaling through tyrosine phosphorylation and interactions with Src family kinases and phosphatases.
CD22 is positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.
